Non-coding antisense RNA can be used to stimulate protein production

Wednesday, October 17, 2012

While studying Parkinson's disease, an international research group made a discovery which can improve industrial protein synthesis for therapeutic use. They managed to understand a novel function of non-protein coding RNA: the protein synthesis activity of coding genes can be enhanced by the activity of the non-coding one called "antisense."

To synthesize proteins, the DNA needs RNA molecules serving as short "transcriptions" of the genetic information. The set of all these RNA molecules is called "transcriptome." In the human transcriptome, along with around 25 thousand sequences of coding RNA (i.e. the sequences involved in the synthesis process), an even larger number of non-coding RNA sequences can be found. Some of these RNAs are called "antisense" because they are complementary to sequences of coding RNA called "sense" (the pairing of a sense and an antisense RNA can be seen as a zip).

The RIKEN Omics Science Center has previously discovered that many of the protein coding genes have corresponding antisense RNAs. A study published in Nature, coordinated by a group of SISSA researchers in Trieste, Italy, has now found that a particular type of antisense RNAs stimulate the translation of the protein coding mRNAs that they overlap to. This is in sharp contrast with the current belief that antisense RNAs are universally associated to negative regulation of protein translation.

Most of the mammalian genome is transcribed producing non-coding RNA. The RIKEN FANTOM projects have earlier demonstrated that the largest output of the genome is constituted by non-coding RNAs. More than 70% of the mRNAs are associated in cells with non-coding antisense RNAs, which are usually thought to negatively repress transcription or translation.

In an exceptional collaborative study based on RIKEN FANTOM sense-antisense cDNA clones, the consortium (including SISSA and the RIKEN Omics Science Center) has found a class of non-coding antisense RNAs that do the contrary of what is currently known: enhance translation of mRNAs with which they are pairing. The researchers identified this function studying the antisense of the mRNA of Uchl1, a mouse gene involved in brain function and neurodegenerative diseases. The team, using bioinformatics and data-mining at RIKEN, has also discovered that the antisense of Uchl1 RNA is not a single case but instead is the representative of a larger class of mammalian antisense RNAs, which function is to increase translation. This is the first report of an antisense RNA that increases protein production, which works both in mouse and human cells and is predicted to have similar function in other organisms.

The mechanism to stimulate translation is based on increased association of mRNAs with ribosomes, which is mediated by a SINEB2 element, a repetitive sequence in the antisense of Uchl1 RNA, which is placed in an inverted orientation in the non-coding RNA. The specificity is given by a short antisense RNA sequence that hybridizes with the initial part of the protein encoding mRNA.

Why is it an important discovery?

Very little used to be known about "long, non-coding" RNAs and this new research sheds light on some of these molecules. "We focused on one gene, Uchl1, whose mutations are linked to some hereditary types of Parkinson's disease," stated Stefano Gustincich, Professor at SISSA. "We have seen that the non-coding antisense RNA matched to this gene is made up of two fragments, the real antisense fragment matching with the sense RNA that codifies the protein and the SineB2 sequence. The antisense fragment has the function of a 'lock' into which the key of the coding RNA specific for that gene is inserted, while the other one has a stimulating function on protein synthesis."

If you change the antisense fragment with the analogous of another gene, the SineB2 sequence maintains its stimulating function on the new gene. "This is important," explained Gustincich "because it means that the action of sineB2 could be used to stimulate protein production for therapeutic use ? any protein ? in industrial synthesis processes."

"We are delighted to see that there is one more function for long non-coding RNAs," says Piero Carninci, Team Leader at RIKEN OSC. "Since the initial discovery that the majority of the genome produces so many non-coding RNAs, there has been a general skepticism related to the possible function of these RNAs. This is a milestone study identifying a novel class of non-coding RNAs which have a key regulatory function, enhancing protein translation. Additionally, this function is mediated by repetitive elements, so far generally considered the 'junk' fraction of the genome, suggesting that the concept that most of the genome is 'junk' should be revisited. After all, there may be function embedded in any part of the genome, which we do not yet understand."

RIKEN and the RIKEN venture company TransSINE Technologies are committed to exploit commercial applications of this specific structure of the antisense RNA.

These RNAs, called SINEUPs, can be engineered to stimulate translation of other proteins, by changing the overlapping antisense region to target any protein of industrial or therapeutic interest. Initial target proteins will include therapeutic proteins, like antibodies or other soluble factors, as well as other basic studies to understand gene function by overexpression of proteins. RIKEN believes that this work can be broadly used.

Beach Cottage girlfriends, you need this recipe in your life.

We all know I am all about the looking good without doing anything much, this is rather tricky with hair and one?s ahem face but, with recipes, this is that in a nutshell.

IMG_9170

This is so ridiculously amazingly easy but tastes like you spent a lot of time thinking, preparing and working at this that it is a Too Easy Recipe all over.

IMG_9206

I don?t have any exciting story to tell you about this?no memories of holidays in the South or indeed the West of France and I have no idea where it came from, when or how I found it but I do know this.

You should make it too.

IMG_9159

This goes beautifully with a barbecue and ticks most admirably the Australian conundrum when one is asked to ?just bring a salad?? I have said before that those 4 humble words when you are fresh off the boat can leave you looking like a prize, English idiot ? your salads need to rock in this land of beauty..this so does that for you quickly and easily

Beach Cottage Recipe

French Carrot Salad

4 large carrots (about 500g)

1 tablespoon parsley

6 tbsp olive oil

1/4 cup lemon juice

1 garlic clove finely chopped or minced

?

1. grate carrots finely

2. whisk lemon juice, olive oil & garlic, salt & pepper

3. pour vinaigrette over carrots and mix

4. serve with the parsley

IMG_9215

THAT IS IT!

IMG_9161

Truly, unbelievably it is as simple as that and it ROCKS.

IMG_9197

This is lazy girl salad with a Beach Cottage Too Easy twist?meaning it tastes fandabbydozy, when all you did was a little bit of grating and pouring of zee oil plus if you, like me, often have sad and lonely looking carrots hanging around in the bottom of your vegetable drawer in the fridge, this my friends, will use them up very well.

IMG_9177

The best thing about this is the Beach Cottage Crew love it?every single time I make it (I double up this recipe) there is never enough and always people asking more?and where veggies and kids are concerned I feel I can?t go wrong with that.

See you next time lovelies.

Boston, MA A new study by Harvard School of Public Health (HSPH) researchers suggests that, for HIV patients receiving highly active antiretroviral therapy (HAART) to treat HIV, there is no benefit from high- vs. standard-dose micronutrient supplementationand that, in fact, high-dose supplements may cause harm. The study is the first large randomized trial to look at how high-dose multivitamin supplementation affects clinical outcomes among people on HAART.

The study appears in the October 17, 2012 issue of the Journal of the American Medical Association (JAMA).

Previous studies have shown that high doses of supplemental micronutrients help HIV patients that are not receiving HAART reduce disease progression and death, thus prolonging the time before HAART initiation is needed. The HSPH researchers wanted to know if high-dose multivitamin supplementation would provide a similar benefit for HIV patients on HAART; although HAART undoubtedly has major benefits, recovery of the immune system is incomplete, and the risks of mortality and opportunistic infections remain high especially in the first few months after HAART initiation.

The researchers, including lead author Sheila Isanaka, research fellow in the HSPH Department of Nutrition, and senior author Wafaie Fawzi, professor of nutrition, epidemiology, and global health and chair of the Department of Global Health and Population at HSPH, studied a group of 3,418 patients with HIV who started HAART between November 2006 and November 2008 in seven clinics in Dar es Salaam, Tanzania. Half of the patients received high doses of supplements including vitamin B complex, vitamin C, and vitamin E; the other half received standard doses at the recommended dietary allowance level for a median duration of 15 months.

The results showed that high-dose supplementation had no effect on several key measures that reveal HIV disease progressionCD4 count, plasma viral load, body mass index, or hemoglobin level concentrationand did not reduce death or disease progression risks for HIV-infected patients. In addition, the researchers found that high doses of multivitamins increased patients' risk of having elevated levels of ALT, an enzyme associated with liver problems and other serious conditions.

"Although the provision of high-dose vitamin supplements has been found safe and efficacious among HIV-infected patients not receiving HAART, the results from this study show that the safety and efficacy of nutritional interventions in the context of potent combination therapies such as HAART need to be further examined," said Fawzi.

"This study provides no clear evidence of a benefit of high-dose micronutrient supplementation compared to standard-dose supplementation in adults receiving HAART, but it highlights the need for further research on how micronutrient supplements can be better positioned alongside antiretroviral drugs to reduce morbidity and mortality due to HIV," said Isanaka.

Micronutrients are key factors in maintaining immune function and neutralizing oxidative stress, and future studies could examine whether micronutrient supplements might be of benefit if they are offered with food, or given in lower doses, or given only after HIV patients have acclimated to HAART therapy, she said.

Thinking about remodeling? Don't Wait! | Arizona Wholesale Supply

?

From The Arizona Republic, Friday, Oct. 13, 2012

After a five-year slump, the home-remodeling industry is showing new signs of life, driven primarily by rising home sales and pent-up demand to make improvements.

Locally, contractors and home-improvement retailers report business is picking up.

National studies also point to growing interest in remodeling. The latest quarterly report of the leading indicator of remodeling activity from the Joint Center for Housing Studies of Harvard University says remodeling activity is poised for double-digit growth by the first quarter of 2013. The center attributed the trend to rising home sales.

If you are a remodeler, this is good news indeed. If you are a consumer, now is the time to ramp up your planning. With business improving, you need to be prepared to take advantage of this timing and act quickly. Designers will get busy. Time and resources become scarce, and the gap doesn't fill overnight. Also, there has already been some upward price pressure on kitchen products and it doesn't appear to be abating. For example, Sub Zero and Wolf have a price increase on November 1st. There's a good chance others will follow in early 2013.

On the other hand, Arizona Wholesale Supply, does have some inventory of popular appliance models, as well as a large closeout department. With three showrooms open to serve you and highly skilled appliance experts to answer your questions, there's no reason to hesitate. Call now to schedule an appointment at the showroom near you:

California city bans smoking in multi-unit housing

By Ronnie Cohen, Reuters

SAN RAFAEL, Calif. - A San Francisco suburb on Monday banned smoking in duplexes, condominiums and other multi-family homes, with city leaders saying they hoped to lead a wave of such regulations across California and ultimately the country.?

The City Council in San Rafael, a community of 57,000 people about 15 miles north of San Francisco, voted unanimously for the ban, following a handful of other California municipalities that have outlawed smoking in buildings with as few as two units.?

"We are happy to blaze a trail," Mayor Gary Phillips said before the vote. "We're most happy to be in the forefront of the issue because we think it will greatly benefit our residents and those visiting San Rafael, and we think it will set the tone for other cities as well."?

Tobacco-control experts predicted that the tough smoking ordinance in San Rafael could touch off a larger movement in other states and cities.?

Stay informed with the latest headlines; sign up for our newsletter

"The San Rafael ban is a very significant event because it will spread," said Robert Proctor, a Stanford University history of science professor. "We're on the downslope of a big curve. Smoking peaked in 1981 with 630 billion cigarettes sold in the United States. Now it's down to 350 billion. And that number will keep on going down until smoking is a distant memory."?

San Rafael is the state's ninth municipality to completely restrict smoking in multi-unit housing, said Pam Granger, advocacy manager for the American Lung Association in California. Granger said California was the only state where local jurisdictions have banned smoking in homes.?

Supporters of the San Rafael measure say it would protect bystanders from the health hazards associated with secondhand smoke, a known carcinogen that can seep through ventilation ducts, doorways and open windows in residential units with shared walls, California Watch reported.?

Although the ordinance has generally been supported by residents who have spoken at city council meetings, the proposal has set the stage for a fierce fight over how far the government should go in regulating peoples' private lives.?

"This proposed smoking ban actually intends to punish people for what they do in their own homes," Thomas Ruppenthal told the city council. "I really feel this is tyranny."?

City officials say they have received roughly 30 emails and calls from residents who say the ban would leave smokers with few places to go, California Watch reported.?

The ordinance also would prohibit smoking on San Rafael's downtown streets - the backdrop for parts of the 1973 film "American Graffiti."?

Watch the most-viewed videos on NBCNews.com?

When George Lucas filmed his coming-of-age movie in his hometown in 1973 people smoked at work, on airplanes, in restaurants, even in schools and hospitals. It was not until 1977 that neighboring Berkeley became the world's first city to restrict smoking in restaurants.?

California restaurants and most workplaces went smoke-free in 1995, and bars, once a smokers' haven, followed in 1998. California and three other states now restrict smoking in cars with children.?

Secondhand smoke kills an estimated 50,000 Americans, including 430 infants, a year, according to a 2006 U.S. Surgeon General's report. It concluded that concentrations of cancer-causing and toxic chemicals might be higher in secondhand smoke than in the fumes that smokers directly inhale.?

California municipalities have used the report to prohibit smoking in apartments and other multi-family homes. In some cases, the laws apply only to new construction or to just a percentage of a housing complex's units.?

But the restrictions have become increasingly strict, and San Rafael's ordinance applies to all homes, new or existing and rented or owned, with shared walls.?

Fitness Tips For Fall - HER Week In Health - Fitness Video by ...

Summer has pretty much come and gone and just because you?ve put your swimsuit or bikini away for the year doesn?t mean it?s time to let yourself go. Hi! I?m Shana O?Connor with EmpowHER.com with another edition of HER Week In Health and I?m here to share with you some great ways to stay in shape for the fall so that you are not fighting another uphill battle the next time bathing suit season comes around.

As you take time to reevaluate your fall schedule, it is important to take a closer look at your physical activity and food choices too. Make several goals for you to reach throughout the season and try and reach each one by Halloween, then Thanksgiving, and then the end of the year. If you do this you will be further ahead of most others who wait until January 1st to get started on a plan.

So just how much exercise should you be doing?

According to the Mayo Clinic, you should be getting ?150 minutes a week of moderate aerobic activity (like walking) OR 75 minutes a week of vigorous aerobic activity (like running or aerobics).?

It?s also important to strength train as well, especially if you are looking to burn fat. Women?s Health Mag.com suggests two 30-minute weight-training sessions a week. Those two sessions a week can reduce overall body fat by about 3 percentage points in just 10 weeks, even if you don?t cut a single calorie.

Women (and men) constantly look to the rich and famous for tips on how to stay thin and beautiful. So it comes as no surprise that celebrities are often in the spotlight when it comes to the new and unusual fitness trends.

Morning show host Kelly Ripa is a big fan of SoulCycle, which puts a new ?spin? on the traditional stationary bike spin class by throwing in positive affirmations throughout the workout to leave you feeling empowered and unstoppable.

FOX Television star Zooey Deschanel enjoys taking Barre [pronounced bar] classes, which is a ballet fusion dance class which aims to create that dancer?s body by using micro-movements and can burn up to 500 calories per 1 hour class.

While doing the same routine as a star might not result in the same body, it can be a way to refresh a stale workout, provide additional motivation, and even burn more calories.

If hitting the gym isn?t your style there is a new study done by the American Heart Association that gives more reason for you to keep moving or get moving.

?Middle-aged adults who regularly engage in leisure-time physical activity for more than a decade may enhance their heart health.?

More than 4,000 people around 50-years old were followed during this study as they kept track of their ?leisure-time physical activities such as brisk walking, vigorous gardening, cycling, playing sports, and doing housework or home maintenance.?

While gardening, housework, or home repair hardly feel like Olympic sports, these activities can still burn a significant amount of calories. According to Fitday.com, you can burn more than 200 calories gardening, while cleaning the house can burn almost 150 calories.

The American Heart Association encouraged doing housework instead of hiring others to do it. But if scrubbing your floor does not sound like fun, walking is a nice way to get started.

No matter what you choose, the important thing is that you get out there and get moving. For more information and workout ideas visit our fitness resource page right here on EmpowHER.com or post your questions in the comments section below. I?m Shana O?Connor for EmpowHER.com and don?t forget to join us right here every week for another edition of HER Week in Health!

Scientists provide window on space radiation hazards

ScienceDaily (Oct. 15, 2012) ? Astrophysicists from the University of New Hampshire's Space Science Center (SSC) have created the first online system for predicting and forecasting the radiation environment in near-Earth, lunar, and Martian space environments. The near real-time tool will provide critical information as preparations are made for potential future manned missions to the moon and Mars.

"If we send human beings back to the moon, and especially if we're able to go to Mars, it will be critical to have a system like this in place to protect astronauts from radiation hazards," says associate professor of physics Nathan Schwadron of the UNH Institute for the Study of Earth, Oceans, and Space (EOS), which houses the SSC.

Schwadron is the lead developer of the new web-based tool known as PREDICCS, which for the first time integrates numerical models of space radiation, a host of real-time measurements being made by satellites currently in space, and "propagation codes" that can accurately project radiation levels out as far as Mars. The tool was made possible through NASA's Living With A Star (LWS) Targeted Research and Technology program.

The website provides updates of the radiation environment on an hourly basis and archives the data weekly, monthly, and yearly. This historical record provides a clear picture of when a safe radiation dose limit is reached for skin or blood-forming organs, for example.

Says Schwadron, "What we really need to know is how hazardous this cycle of radiation is. How often do we see large events that have significant risk associated with them? Those questions can only be answered if you're continually building up the database of events and the risk associated with them."

He notes further that the space science community has traditionally viewed radiation hazards in space as a "showstopper" and that until PREDICCS there has never been an extremely accurate, nearly real-time means of challenging that.

"There hasn't been enough work done to ask, 'Is it really a showstopper and, if so, why, and what are the problems we need to solve so that it isn't a showstopper?'"

That work has now been done, and the proof is in PREDICCS.

Among other satellite measurements used by PREDICCS are solar energetic particle data from the Cosmic Ray Telescope for the Effects of Radiation (CRaTER) instrument on NASA's Lunar Reconnaissance Orbiter. CRaTER, whose principal investigator is Harlan Spence, director of EOS and a co-developer of PREDICCS, has made the most accurate and comprehensive measurements of radiation at the moon since the dawn of the space age.

During several recent large solar events in which the sun, waking from an unusually long quiet period, sent billions of tons of high-energy particles rippling through space, the radiation levels measured by CRaTER as it orbited the moon were matched almost perfectly by PREDICCS.

"For the whopping solar events of January 23 and March 27 of this year, our predictions seem to be within 20 to 30 percent of what was observed, which is incredible. These types of highly accurate comparisons have never been made before," Schwadron says.

CRaTER, which gauges radiation doses using a high-tech material called "tissue-equivalent plastic" that mimics human muscle, has thus not only provided the validation that PREDICCS models are accurate, but has done so in the context of how the radiation data would impact human beings on the moon or on a mission to Mars.

"We needed to accurately assess what the biological impacts are to make the best quantitative comparisons between models and observations," says Schwadron, "and having a system like this in place now is sort of like flying a trial balloon in preparation for a return to the moon and a trip to Mars."

To make its radiation assessments, PREDICCS integrates two radiation environment models, including the Earth-Moon-Mars Radiation Environment Module (EMMREM) developed at UNH.

"Complex applications like EMMREM are able to leverage observations from all relevant space missions," notes NASA's Madhulika Guhathakurta, LWS program scientist.

A blog has been developed for PREDICCS that allows people to understand how the tool works and how to interpret the various graphs of radiation dosage.

Note's Schwadron, "For the first time people are able to see the affects of space radiation playing out in near real-time, and this opens a new window to an otherwise invisible world."

To view the PREDICCS website, visit http://prediccs.sr.unh.edu/.

The goal of the LWS is to develop the scientific understanding needed for the U.S. to effectively address those aspects of the connected sun-Earth system that may affect life and society. The program's Targeted Research and Technology objectives can be achieved by data analysis, theory and modeling, and the development of tools and methods.

Thousands rally for Malala, girl shot by Taliban

As doctors debated whether to send Malala Yousafzai abroad for care, thousands rallied in her name, including hundreds of schoolgirls who gathered in Afghanistan. NBC's Amna Nawaz reports.

By NBC News and news services

KARACHI, Pakistan -- Tens of thousands rallied in Pakistan's largest city Sunday in support of a 14-year-old girl who was shot and critically wounded by the Taliban for promoting girls' education and criticizing the militant group.?

The demonstration in the southern city of Karachi was by far the largest since Malala Yousufzai and two of her classmates were shot on Oct. 9 while returning home from school in Pakistan's northwest.?

The attack horrified people inside and outside Pakistan and sparked hope among some that it would prompt the government to intensify its fight against the Taliban and their allies.?

But protests against the shooting have been relatively small until now, usually attracting no more than a few hundred people. That response pales in comparison to the tens of thousands of people who held violent protests in Pakistan last month against a film produced in the United States that denigrated Islam's Prophet Muhammad.?

Demonstrations in support of Yousufzai ? and against rampant militant violence in the country in general ? have also been fairly small compared with those focused on issues such as U.S. drone attacks and the NATO supply route to Afghanistan that runs through Pakistan.?

A Pakistani woman shouts slogans during a protest march against the assassination attempt by the Taliban on 14-year-old activist Malala Yousafzai, in Karachi on Sunday. Malala is making "slow and steady progress" in her recovery, the military said.

Right-wing Islamic parties and organizations in Pakistan that regularly pull thousands of supporters into the streets to protest against the U.S. have less of an incentive to speak out against the Taliban, who share their desire to impose Islamic law in the country ? even if they may disagree with some of the militant group's violent tactics.?

Pakistan's mainstream political parties are often also more willing to harangue the U.S. than direct their people power against Islamist militants shedding blood across the country ? partly out of fear and partly because they rely on Islamist parties for electoral support.?

One of the exceptions is the political party that organized Sunday's rally in Karachi, the Muttahida Quami Movement. The party's chief, Altaf Hussain, criticized both Islamic and other mainstream political parties for failing to organize rallies to protest the attack against Yousufzai.?

Positive developments have been reported regarding the recovery of 14-year-old Malala Yousufzai, who was shot by the Taliban near her school on Tuesday. NBC'S Amna Nawaz reports.

He called the Taliban gunmen who shot the girl "beasts" and said the shooting was an attack on "the ideology of Pakistan."?

"Malala Yousufzai is a beacon of knowledge. She is the daughter of the nation," Hussain told the audience by telephone from London, where he is in self-imposed exile because of legal cases pending against him in Pakistan. His party is the strongest in Karachi.?

Watch World News videos on NBCNews.com

Many of the demonstrators carried the young girl's picture and banners praising her bravery and expressing solidarity.?

Yousufzai earned the enmity of the Taliban for publicizing their behavior when they took over the northwestern Swat Valley, where she lived, and for speaking about the importance of education for girls.?

The group first started to exert its influence in Swat in 2007 and quickly extended its reach to much of the valley by the next year. They set about imposing their will on residents by forcing men to grow beards, preventing women from going to the market and blowing up many schools ? the majority for girls.

Images of daily life, political pursuits, religious rites and deadly violence.

Yousufzai wrote about these practices in a journal for the BBC under a pseudonym when she was just 11. After the Taliban were pushed out of the valley in 2009 by the Pakistani military, she became even more outspoken in advocating for girls' education. She appeared frequently in the media and was given one of the country's highest honors for civilians for her bravery.

The Pakistani Taliban said they carried out the shooting because Yousufzai was promoting "Western thinking." Police have arrested at least three suspects in connection with the attack, but the two gunmen who carried out the shooting remain at large.

The young girl was shot in the neck, and the bullet headed toward her spine. Doctors at a military hospital operated on her to remove the bullet from her neck, and she was put on a ventilator. Her condition improved somewhat on Saturday when she was able to move her legs and hands after her sedatives were reduced.

On Sunday, she was successfully taken off the ventilator for a short period and later reconnected to avoid fatigue, the military said. Doctors said she is making slow and steady progress.

The possibility of transferring Malala overseas for continued treatment was still being considered, a military spokesman said. Pakistan has arranged a specially equipped air ambulance with the United Arab Emirates that will be used if doctors decide to move her abroad.

Visas are being finalized for the air ambulance crew and six doctors who will accompany the flight, the Pakistani ambassador to the country, Jamil Ahmed Khan, told Pakistan's Geo TV. Arrangements have been made to treat the girl at three hospitals in Dubai and Abu Dhabi, he said.?

The UAE Embassy in Islamabad could not immediately be reached for comment.?

No decision has yet been taken to send the girl abroad, but the air ambulance is part of the contingency plan, the Pakistani military said.?

Afghan President Hamid Karzai has written letters to top political and religious leaders in Pakistan denouncing the attack on Yousufzai and asking them to help battle extremism in both countries, the president's office said in a statement issued late Saturday. Karzai wrote that he views the shooting as an attack on Afghanistan's girls as well.

How specify platform independent paths in java.security and policy ...

I want to ship a single security policy file to our customers - for both Linux and Windows. In the java.security file, I add a line to point to our file, but due to path differences I have to specify it differently for Windows and Linux

policy.url.3=file:/${our.home}/lib/OurSecurity.policy  # Windows policy.url.3=file:${our.home}/lib/OurSecurity.policy   # Linux

That first "/" after the "file:" is needed on windows since the variable our.home is defined as "C:\foo". I have the same problem in the policy file itself:

grant codeBase "file:/${our.home}/-" // Windows grant codeBase "file:${our.home}/-" // Linux

I'm especially looking for a clever way to specify the codeBase in a platform independent way. By the way, redefining our.home is not really an option, as it's needed in a FilePermission clause.

US deficit tops $1 trillion for fourth year

Graphic shows the U.S. budget deficit

Graphic shows the U.S. budget deficit

(AP) ? The U.S. budget deficit has topped $1 trillion for a fourth straight year, but a modest improvement in economic growth helped narrow the gap by $207 billion compared with last year.

The Treasury Department said Friday the deficit for the 2012 budget year totaled $1.1 trillion. Tax revenue rose 6.4 percent from last year to more than $2.4 trillion, helping contain the deficit.

The government's revenue rose as more people got jobs and received income. Corporations also contributed more tax revenue than in 2011.

Government spending fell 1.7 percent to $3.5 trillion. The decline reflected, in part, less defense spending as U.S. military involvement in Iraq was winding down.

Barack Obama's presidency has now coincided with four straight $1 trillion-plus annual budget deficits ? the first in history and an issue in an election campaign that ends in Nov. 6.

Obama's Republican challenger, Mitt Romney, contends that Obama failed to achieve a pledge to halve the deficit he inherited by the end of his first term.

When Obama took office in January 2009, the Congressional Budget Office forecast that the deficit for that year would total $1.2 trillion. It ended up at a record $1.41 trillion.

The increase was due, in part, to higher government spending to fight the worst recession since the Great Depression of the 1930s Tax cuts enacted under President George W. Bush and wars in Iraq and Afghanistan contributed to the deficits.

The budget gaps in 2010 and 2011 were slightly lower than the 2009 deficit as a gradually strengthening economy generated more tax revenue. But the deficits still exceeded $1 trillion.

Obama is campaigning for a second term with a pledge to cut deficits by $4 trillion over the next decade. He says he would do so by ending the Bush-era income tax cuts for higher-income Americans and by restraining the growth of spending.

Romney has said he would cut spending growth to help narrow the budget gap. He would cap spending at 20 percent of the economy by 2016. Spending in 2012 accounted for about 23 percent of the economy.

The government borrowed about 31 cents of every dollar it spent in 2012. The string of $1 trillion-plus deficits has driven the national debt above $16 trillion. The magnitude of that figure has intensified debate in Congress over spending and taxes but little movement toward compromise.

Many fear the budget deadlock will send the economy over a "fiscal cliff" next year, when tax increases and deep spending cuts will take effect unless a budget deal is reached.

Obama wants to eliminate the income tax cuts for families that make more than $250,000.

Republicans in Congress and Romney have resisted. They argue that with the economy still weak, the government should not be raising anyone's taxes.

Congress may address the budget crisis during a lame-duck session of Congress after the November elections.

Perfect Health and Fitness Provides the Key to a Life Without - SBWire

Bear River, Nova Scotia -- (SBWIRE) -- 10/12/2012 -- According to the National Institute of Health, in 2004, 104.7 outpatient visits were made involving a digestive disease. Of these visits, 13.5 million resulted in a hospitalization and 236,164 deaths were attributed to a digestive disorder. All age groups were seen during these visits. although the highest rate was for those age 65 or older. The total cost for these visits was $141.8 billion, with $97.8 billion of that being for direct medical costs. With rising health care costs, this should be of concern to every American, states Elke Neher, founder of Perfect Health and Fitness.

Digestive diseases encompass a number of medical conditions, including, but not limited to, abdominal wall hernias, gastritis, inflammatory bowel disease and viral hepatitis. "When someone has been diagnosed with a digestive disease of any type, he or she may immediately turn to the Internet for information. Sherry Brescia, creator of "Great Taste No Pain", did the same thing when she suffered from severe pains in the past. Sadly, this is often the worst thing to do," Ms. Neher says.

"As an expert health researcher, Sherry Brescia works with patients suffering from digestive diseases on a regular basis. When she began her research, she quickly realized that much of the information provided online was actually incorrect. This caused her great concern as many are inclined to believe what they read on the Internet is fact," Ms. Neher continues.

As a result of this discovery, Ms. Brescia chose to write "Great Taste No Pain". Although there was a great deal of skepticism about her site when it first started," Ms. Neher explains, "Over time, people began to realize that the information provided in the books is trustworthy and accurate. As a result, customers are pleased with the results obtained and come back satisfied. No matter what digestive disorder one is suffering from, the information provided by Ms. Brescia is of great help. Be sure to visit her site to learn more about her work." Ms. Brescia offers a 60 day money back guarantee. If customers are not satisfied, the books may be returned for a full refund.

"At Perfect Health and Fitness, we offer advice on a wide range of digestive issues. Request Sherry Brescia's free report Pain-Free in 1 Day to get started on the path to a life free of digestive diseases," Ms. Neher goes on to say. "Give us a try today and see how we can be of help. No matter what digestive issues patients are suffering from, we offer information to assist them in resolving the issues once and for all."

DLink PowerLine AV Mini Adapter kit keeps wired networks as subtle and cheap as possible

D-Link still sees there being a place for powerline networking, such as a basement home theater, but knows that many of us don't want an overly clunky set of adapters or to effectively buy a second network. Its new (if slightly awkwardly named) PowerLine AV+ Mini Adapter Starter Kit caters just to that desire for a no-hassle network in places WiFi won't reach. The bundle carries a pair of small, 200Mbps adapters that won't monopolize the wall outlets, a pair of Ethernet cables to link up... and that's it. Apart from the optional 128-bit AES encryption, there's nothing else needed to either make a direct connection or graft powerline technology into an existing network. At $60, the already shipping kit is also just low enough in price that there's very little standing between us and a reliable connection for a console or placeshifter.

LG Nexus Leaked Photos - Business Insider

We must be getting close to Google's new Nexus smartphone launch, because we're seeing new leaks every day.

The latest photos come from a Belarus-based site called Onliner, which we found on The Verge. It seems like they got their hands on Google's new Nexus phone a bit early. And these are the clearest photos of the new Nexus we've seen yet.

So far, we know the new Nexus will be made by LG and feature some very impressive specs under the hood. There's also a possibility other manufacturers like Samsung, HTC, and Sony will make Nexus-branded devices in time for the holidays.

We included a few photos of the LG Nexus below. You can head to Onliner for even more >

Walking for Cancer : Ideas For Women Health and Fitness Blog

Cancer research and funding is at the forefront of most media outlets especially breast cancer research and funding. It does my heart good as a woman to see NFL players wearing pink shoes or NBA players wearing pink to show solidarity for women?s health issues. Women can do the same service for men with cancers such as testicular, colon, or lung cancers and we should.

September was Childhood cancer awareness month and October already sports breast cancer awareness. Cancer awareness shouldn?t have only one month to make people aware. Walking for cancer research and funding can be done almost any time of the year and walking is a great exercise for those who are making a step towards better health.

The Leukemia and Lymphoma society often have Night Walks to make those aware of their needs and their wonderful advancements in research and treatments. Visit www.lls.org for tons of good information and resources for children and adults with leukemia, lymphoma, or other blood cancers. A quick Internet search can also offer you pages of links to specific cancers, lists of symptoms or warning signs for cancers, and ways to volunteer and contribute financially to cancer research.

Taking an interest in cancer research and funding those programs that are working so hard for the cure can be a win-win for each of us. Individuals can start their own walking group to raise awareness or to raise funds for someone in their community who is dealing with cancer. Each of us can be responsible for reaching out to the American Cancer Society in our own areas to see what we can do to help. And any time you can walk in a cancer walk-a-thon to support research and a cure, you are promoting your own health with those steps. Together we can make a difference in this dreaded disease if we start right now. At the very least read up on the warning signs and get your appointment made to have a physical. It can be important to your life.

WellnessFX Practitioner Ben Greenfield on Training and Winning ...

Depending on your health hurdles, your triumphs and any lofty fitness goals you may have, your health guru may be your family physician, your acupuncturist or your personal trainer. At WellnessFX, we offer a network of practitioners that range from medical doctors to dieticians and nutritionists, all with a variety of backgrounds and specialties.

One of our wellness gurus is Ben Greenfield, a WFX practitioner, sports nutritionist and personal trainer. Ben helps his clients with weight control, sports performance, holistic medicine and lifestyle management. A triathlete himself for the last nine years, Ben demonstrates a devotion to health, quantified living and the victory that comes with working toward your goals.

Using a tailored training and nutrition program, much like the ones he recommends for his clients, Ben recently finished first at the Leadman 125 Triathlon in Bend, Oregon. The gorgeous Pacific Northwest course starts with a 2.5km swim in chilly Citrus Lake at about 5,000 feet, then transitions into a hilly 106km bike course, and ends with a 16.5km run along the banks of the Deschutes River.

Ben?s unique style of training results in an eight to 10 hour training week, in contrast to most tri-trainers who push training to an average 15 to 25 hours per week. ?I only train for 60 minutes each day, with one long workout on the weekends,? he explains. ?Rather than training excessively, I put more focus into the quality and intensity of my training, recovery, sleep and diet.

?As a coach, I have the challenge of looking at my client?s healthy history, exercise history, goals and limitations, and custom designing a training and nutrition plan accordingly,? Ben describes. While training and nutrition needs vary among Ben?s clients, the overlying philosophy stays the same: ?Quality training over quantity of training, the elimination of ?junk miles? and a focus on the variables that many athletes tend to put on the backburner.? These, according to Ben, include holistic, nutrient-dense fueling, smart recovery, stress control and deep sleep.

?Many of the athletes I coach (and myself) also implement alternative methods such as cold thermogenesis, electro stimulation and customized nutrition supplement plans. All of which make a big difference.?

Check out Ben?s full race report here and his approach to a winning race-day nutrition protocol.

Mysterious Android tablet shows up in North Korea

North Korean tablet

Yes, you read that correctly; a tablet was spotted presumably running Android in North Korea's capital of Pyongyang. The photo was taken during a trade fair where various approved electronic were being shown off.

Due to the closed nature of North Korea, we don't know much about the tablet. No hardware specs, no software information and certainly no dates for availability. One of the only tidbits that was discovered was a box in the background that read Samjiyo, which is the name of another North Korean tablet introduced in June.

From the photo, it looks to have around a 10-inch screen and has some applications installed. Other than that, it's anybody's guess. If we were to surmise about a North Korean tablet, we'd assume it's extremely locked down and will not run Google's core services. Remember North Koreans are prohibited access to the wider Internet. Interestingly enough, it seems as though the Seoul Google office took interest in this photo as well and are trying to find out more information, just as we are.

There's a certain irony in a closed nation like North Korea being able to develop its own tablet because of an open-source OS like Android, don'tcha think?
